PCR Inhibitor Tolerance

Superior PCR inhibitor tolerance with SsoAdvanced™ universal SYBR® Green supermix. An ADAR qPCR assay with an input of 5 ng cDNA was performed with 10, 20, and 40% Eagle’s minimum essential medium supplemented with 10% fetal bovine serum spiked in the cDNA to act as an inhibitor. GC-Rich and AT-Rich qPCR

Optimal qPCR efficiency and sensitivity with SsoAdvanced™ universal SYBR® Green supermix. qPCR reactions were performed on a CFX96™ real-time PCR detection system. A, a 61% GC-rich TIP assay with an input of 50 ng–5 pg gDNA produced standard curve R2 = 0.997, efficiency = 102%, and slope = –3.27; B, a 71% AT-rich ADAR assay with an input of 10 ng–10 pg cDNA produced standard curve R2 = 0.999, efficiency = 92%, and slope = –3.52. PrimePCR™ Assay Compatibility

SsoAdvanced™ universal SYBR® Green supermix is a high-performance real-time PCR supermix based on Bio-Rad's patented* Sso7d fusion protein technology. This supermix is formulated for a wide range of real-time PCR applications and for use with all ROX dependent or independent real-time PCR systems. The dsDNA binding protein, Sso7d, stabilizes the polymerase-template complex, providing superior inhibitor tolerance, increased processivity, specificity, and greater speed without affecting PCR sensitivity, efficiency, or reproducibility.
