PrimePCR™ Digital Assays – Performance Data

PrimePCR digital assays are predesigned, lab-validated assays. The following data illustrate the performance of the mutation detection and copy number variation assays:

Mutation Detection Assays
Mutation detection assays on the ddPCR™ platform enable detection of 0.001% mutant fractions because partitioning increases sensitivity by isolating the target signal from competing background.

High sensitivity of PrimePCR™ ddPCR™ mutation detection assays provides precise quantification of JAK2 V617F mutation in the presence of wild-type DNA.

Mutation Detection Assays

A, 2-D fluorescence amplitude plot shows duplicate wells of a mixed mutant:wild-type sample across multiple percentage mutant blends. The black cluster on the plot represents the negative droplets, the green cluster represents the droplets that are positive for the wild-type DNA only, the blue cluster represents the droplets that are positive for the mutant DNA only, and the orange cluster represents the droplets that are positive for both wild-type and mutant targets;. B, 2-D fluorescence amplitude plot shows duplicate wells of a wild-type only sample; C, fractional abundance plot shows the percentage frequency (orange markers) of the mutant DNA in a wild-type DNA background. Copy Number Variation Assays
PrimePCR ddPCR copy number assays provide superior resolution and precision. The massive partitioning of a copy number variation ddPCR reaction across up to 20,000 droplets enables the discrimination required to determine small fold changes.

Our digital PCR assays are hydrolysis probe-based assays that were designed by experts in the digital PCR field. Every digital PCR assay has been experimentally validated to ensure optimal performance for its target application:

  • Copy number variation (CNV) analysis — determine small changes in copy number without the use of a standard curve
  • Mutation detection — detect one mutant in a background of at least 2,000 wild-type molecules

These predesigned assays for digtal PCR come with a FAM or HEX fluorophore.
