Overview
How much of your favorite foods are genetically modified?
Use the GMO Investigator Real-Time PCR Starter Kit to teach students the principles and techniques of real-time quantitative polymerase chain reaction (qPCR) within the context of testing food for the presence of genetic modifications. Students extract DNA from food items, then use real-time quantitative polymerase chain reaction (qPCR) to test for the presence of DNA sequences that are present in >85% of genetically modified (GM) crops: the 35S promoter of the cauliflower mosaic virus and the terminator of the nopaline synthase gene of Agrobacterium tumefaciens. Quantitate and determine what fraction of the food product is made with genetically modified ingredients, in the same manner as standard testing laboratories do!
The GMO Investigator Real-Time PCR Starter Kit combines the components of the GMO Investigator Kit with the reagents and plastics needed to perform the same activity using qPCR. Students analyze their favorite foods as they:
- Learn the basics of qPCR analysis as they determine the presence or absence of GMO sequences in their choice of test food
- Conduct sophisticated scientific procedures with multiple levels of controls to assess the validity of the results
- Investigate how to perform melt-curve analysis and quantitation through use of a standard curve, all within the context of food testing and labeling
Use of this kit requires the use of real-time PCR system, such as the CFX Duet Real-Time PCR System for the Classroom.
Activities Include
- Use real-time PCR to analyze a set of dilutions of positive control DNA (compare primer sets, use a standard curve for quantitation, perform a melt-curve analysis)
- Extract DNA from food samples
- Perform real-time PCR of DNA from food samples
- Post-lab discussion and debate activity
- Optional: compare results with those obtained by traditional PCR (the GMO Investigator Kit is included as a component of this kit)
Features and Benefits
- Three-session laboratory activity, 45 min per session
- Learn the molecular basis of DNA amplification reactions using real-time qPCR detection systems, all within a real-world application
- Explore the impacts of pipetting accuracy and reliability on qPCR results
