Overview

Crystal Universal Reporters generate fluorescent signals when activated during a digital PCR (dPCR) reaction using target-specific Crystal Flex Probe detection chemistry, allowing for specific detection and quantification of multiple targets within a single reaction.

Crystal Universal Reporter 7 is supplied as ready-to-use 40x reporter solutions (Tube A and Tube B), configured for 200 reactions, for detection across multiple fluorescence channels on naica Prism3, Prism6, and Nio Systems. Tube A supports detection in the blue, green, and red channels. Tube B supports detection in the teal, yellow, and infrared channels, and additionally enables long-shift channel detection on Nio Systems.

Their universal design allows them to be used with any Crystal Digital PCR™ assay, providing a modular approach to detection. The same set of Crystal Universal Reporters can be used to detect and quantify all targets of interest, regardless of the specific assay or degree of multiplexing.

Crystal Flex Probes are non-fluorescent oligonucleotides specific to the target of interest. Each probe contains a non-target-specific sequence that acts as a tag oligonucleotide, triggering fluorescence in the complementary Crystal Universal Reporter when released during PCR. This unique color combination, high-plex assay technology with Crystal Universal Reporters makes detecting more than seven targets individually accessible, with a streamlined 1D thresholding analysis.

Color combination is used in Crystal Digital PCR assays when higher-order multiplexing is required to detect more targets than the number of fluorescent channels available in a single dPCR reaction. A wide variety of catalog assays are available for oncology applications, including but not limited to point mutation and promoter methylation detection and quantification.

Crystal Universal Reporter 3 is designed for Crystal Digital PCR assays with up to three fluorescence colors.

Crystal Universal Reporter 7 supports high multiplexing beyond three colors and up to 21-plex with color combination technology.

For Crystal Digital PCR assays with multiplex levels up to three colors, only Crystal Universal Reporter 3 (Tube A) is required. For higher multiplex levels, Crystal Universal Reporter 7 (Tube A and Tube B) is required.
