CRISPR Gene Editing Is More Than Just a Cut

CRISPR is hailed as a revolutionary tool used by scientists to rewrite DNA and tackle issues like disease-causing mutations head-on. Yet, while the CRISPR-Cas9 system often takes the spotlight for its precise DNA cutting, it's just the opening act. The cell's DNA repair machinery must also step in to turn those cuts into desired and transformative genetic edits.

Where other educational kits merely showcase the cutting function of CRISPR-Cas9, the Bio-Rad Out of the Blue Gene Editing Kit demonstrates the need for both cutting and repair in gene editing. Students utilize CRISPR-Cas9 alongside homology-directed repair (HDR) to both cut and insert a stop codon into the chromosomal lacZ gene of E. coli. They can then use PCR to confirm that an edit was made.

The carefully designed bacterial strain and plasmids in the Out of the Blue Gene Editing Kit allow students to safely explore and learn about the components of CRISPR gene editing, all with minimal equipment required. Clearly visible results allow students to see what happens if the Cas9 enzyme does not have a guide RNA to direct a cut, and students also witness the vital role of DNA repair machinery in the gene editing process.

In the time it takes to perform a bacterial heat shock transformation, students perform true CRISPR-Cas9 genome editing to change a gene in a living cell, as is done in real-world applications of the technology!
