Overview
Use Kovacs Reagent for the detection of bacteria, such as coliforms including E. coli, that under aerobic conditions are able to split tryptophan into indole and α-aminopropionic acid. The reagent contains para-dimethylaminobenzaldehyde (DMAB), and concentrated hydrochloric acid. After growth in a tryptophan-containing broth such as Schubert Broth (3569634) or BGBLB (Brilliant Green Bile Lactose Broth) (3578024), addition of Kovacs Reagent results in an immediate local red color in indole-producing cultures, due to DMAB complexing with indole.
