Vector Backbone Assays

Bio-Rad’s ddPCR Cell and Gene Therapy (CGT) Assays are custom ddPCR-optimized assays generated by the Cell and Gene Therapy Design Engine for your target Gene of Interest. These assays offer the ease and accuracy of ddPCR technology and come in multiple fluorophores to enable multiplexed Cell and Gene Therapy applications, including Viral Titer and Vector Copy Number measurements. Use the ddPCR CGT Assays individually or multiplex them with the Vector Backbone Assays to precisely measure the concentration of the therapeutic gene in your sample.

ddPCR Cell and Gene Therapy Assays provide superior resolution and precision. A, 2-D fluorescence amplitude plot shows six replicate wells of an AAV sample (virus dilution 2) assayed on the QX200 AutoDG ddPCR System with the AAV ITR-2 Assay in FAM (ddPCR EXD Assay: dEXD15274642), duplexed with a custom Gene of Interest (GOI) Assay in HEX (ddPCR CGT Assay), and MspI as the compatible restriction enzyme. The black cluster on the plot represents negative droplets, the blue cluster represents droplets that are positive for AAV ITR-2 only, the green cluster represents droplets that are positive for GOI only, and the orange cluster represents droplets that are positive for both AAV ITR-2 and GOI targets. B, Concentration plot shows the average AAV ITR-2 and GOI target concentrations for three AAV sample dilutions and No-template Control (NTC). Note the consistent AAV ITR-2:GOI ratio, which can be used as an indicator of AAV packaging efficiency. ddPCR Cell and Gene Therapy are provided in a 20X ready-to-use primer-probe mix, available in six fluorophore choices, optimized for use with ddPCR Supermix for Probes (No dUTP) or ddPCR Multiplex Supermix.
