Exclusive Sso7d Fusion Polymerase Technology to Empower Your PCR

Sso7d, a 7 kD protein obtained from Sulfolobus solfataricus, binds to dsDNA without any sequence specificity.

Sso7d is covalently linked to a DNA polymerase and is able to stabilize the polymerase-template complex without compromising the structural integrity, thermal stability, or catalytic activity of the enzyme.

The unique structure of this Sso7d fusion polymerase provides several significant advantages over ordinary DNA polymerase:

  • Increased processivity
  • Effective amplification in GC-rich regions or strong secondary structure
  • Efficient amplification of longer products
  • Superior tolerance to PCR inhibitors

PCR Inhibitor Tolerance

Superior PCR inhibitor tolerance with SsoAdvanced™ Universal Probes Supermix.

A GAPDH qPCR assay with an input of 50 ng cDNA was performed with 1, 2, 2.5, and 3 µM hematin spiked in the cDNA to evaluate tolerance. The SsoAdvanced Universal Probes Supermix tolerated greater levels of inhibition compared to the competitor mix. Cq, quantification cycle.



Duplex Reaction

Superior qPCR performance in a duplex reaction with SsoAdvanced™ Universal Probes Supermix.

Predesigned TaqMan gene expression assays in a duplex reaction with 10 ng–100 fg cDNA, using an 18S rRNA assay (), and 100 ng–10 pg cDNA, using a TFRC assay (), were performed on an Applied Biosystems ViiA 7 system. 18S rRNA efficiency = 98%; TFRC efficiency = 101%. GC-Rich and AT-Rich qPCR

Optimal qPCR efficiency and sensitivity with SsoAdvanced™ Universal Probes Supermix.

A 72% GC-rich CBP assay, with an input of 100 ng–10 pg cDNA, was performed on a CFX96™ Real-Time PCR Detection System and produced standard curve R2 = 0.997 and efficiency = 97%.



A 71% AT-rich APC assay, with an input of 500 ng–50 pg gDNA, was performed on an Applied Biosystems 7500 system and produced standard curve R2 = 0.998 and efficiency = 98%. RFU, relative fluorescence units; ΔRn, baseline-corrected normalized reporter.
