iTaq™ SYBR® Green supermix with ROX gives superior results on the ABI PRISM 7000 sequence detection system. The Bio-Rad supermix (green) and other ROX-containing supermixes (red and blue) were used to amplify tenfold serial dilutions (107 to 10 copies) of the GAPDH gene in a plasmid template. Reactions were performed in quadruplicate (replicates not shown). The Bio-Rad supermix consistently generated earlier threshold cycle (CT) values than the other mixes, and produced high delta Rn values.

Six Orders of Magnitude

iTaq™ SYBR® Green supermix with ROX performs well over six orders of magnitude on the ABI PRISM 7700 sequence detection system. The Bio-Rad supermix was used to amplify tenfold serial dilutions (107 to 10 copies) of the β-actin gene in a plasmid template. Reactions were performed in quadruplicate. Standard curve had r = 0.996, slope = –3.40, efficiency = 96.8%.

Uniform Results

iTaq™ SYBR® Green supermix with ROX produces highly uniform results on the ABI PRISM 7000 sequence detection system. The Bio-Rad supermix was compared to supplier A's ROX-containing supermix by amplifying 102 copies of the β-actin gene from a plasmid template (48 replicates per mix). The Bio-Rad supermix generated more uniform results with earlier threshold cycle (CT) values (mean CT = 24.4, SD = 0.229) than the alternate supplier's ROX-containing supermix (mean CT = 28.0, SD = 0.433). All reactions were performed in the same 96-well plate.

iTaq™ SYBR® Green supermix with ROX is a 2x ready-to-use reaction mix, formulated to deliver maximum PCR efficiency and sensitivity and a robust fluorescent signal in real-time qPCR assays. This formula yields high performance with cDNA as well as genomic and plasmid DNA templates over a broad dynamic range, achieving sensitive and specific amplification over at least six orders of magnitude. This supermix is preblended with hot-start iTaq DNA polymerase, a proprietary buffer and stabilizer, nucleotides, an optimized SYBR® Green I dye that minimizes the potential for primer-dimer formation, and ROX passive reference dye to correct for interwell signal variation. It has been validated for use on all ROX-dependent optical thermal cyclers and with any real-time detection chemistry.
