Relative Focusing Power

Relative Focusing Power of IPG Strips

Relative focusing power expresses the enhanced resolution expected in the first dimension when using IPG strips of different lengths or pH ranges. The 7 cm pH 3–10 IPG strip is arbitrarily assigned a baseline focusing power of 1.0 in order to calculate the relative focusing powers of the other strips.

The calculations are done as follows:

When two strips have the same pH range but differ in length — the ratio of the strip lengths gives the relative focusing power of the longer strip. Thus, compared to a 7 cm pH 3–10 strip, the 11 cm pH 3–10 strip has a relative focusing power of 11 cm/7 cm = 1.6
When two strips have the same length but differ in pH range — the ratio of the pH ranges (in number of pH units) gives the relative focusing power of the strip with the smaller range. Thus, compared to a 7 cm pH 3–10 strip, the 7 cm pH 5–8 strip has a relative focusing power of (7 pH units)/(3 pH units) = 2.3

Narrow-range strips provide an increase in resolution. As illustrated in figures A–D below, three overlapping narrow-range strips can dramatically expand the region for analysis. This narrow-range advantage is illustrated with 7, 11, and 7 cm strips below but applies to 18 and 24 cm strips as well.

Initial screening of complex samples is simplified. Figures A–C, series of overlapping narrow pH-range ReadyStrip™ IPG strips enhance and increase resolution by creating overlapping data equivalent to 40 cm of pI separation across the entire pH 3–10 range (Figure D). In combination with the ReadyPrep sequential extraction kit for fractionating samples by solubility, this significantly increases your resolving power and allows definition of proteins not visible by traditional methods. Micro-Range Strips

Micro-range IPG strips enable the highest resolution available for IPG strips. The micro-range strips cover as small a range as 1 pH unit. These are particularly well suited for resolving different isoforms of the same protein.

E. coli lysate sample from the ReadyPrep™ 2-D starter kit separated on Bio-Rad's micro-range IPG strips on 8–16% handcast gels in the PROTEAN® Plus Dodeca™ cell.

Increased Resolving Power

Increase in resolving power in the first dimension with overlapping pH range IPG strips. E coli lysate (40 µl per gel) was run on 11 cm overlapping narrow pH range ReadyStrip IPG strips and focused for 20,000 V-hr. The strips were then transferred to 8–16% Tris-HCI Criterion precast gels for the second dimension run. The gels were strained with colloidal Coomassie Blue stain. More proteins were detected on the three overlapping gels than on the single 3–10 pH range gel. Note the improved resolution of proteins in the circled areas.

IPG strips simplify first-dimension separations by immobilizing the pH gradient on an easy-to-handle support strip. ReadyStrip™ IPG strips are cast using high-purity IPG monomers and are thoroughly tested for quality and performance to deliver gel-to-gel reproducibility. ReadyStrip IPG strips, available in a wide selection of pH gradients and strip lengths from 7 to 24 cm, fit Bio-Rad vertical electrophoresis cells and gels.

Gradient Selection

Micro-range gradients for the ultimate in resolution — zoom in on an area of interest or fine-map an entire proteome; strips are carefully designed with sufficient overlap to allow spot matching while limiting the extent of redundant data
