Overview

Exclusive Sso7d Fusion Polymerase Technology to Empower Your PCR

Sso7d, a 7 kD protein obtained from Sulfolobus solfataricus, binds to dsDNA without any sequence specificity.

Sso7d is covalently linked to a DNA polymerase and is able to stabilize the polymerase-template complex without compromising the structural integrity, thermal stability, or catalytic activity of the enzyme.

The unique structure of this Sso7d fusion polymerase provides several significant advantages over ordinary DNA polymerase:

  • Increased processivity
  • Effective amplification in GC-rich regions or strong secondary structure
  • Efficient amplification of longer products
  • Superior tolerance to PCR inhibitors

Superior qPCR Performance in the Presence of an Inhibitor

An ADAR qPCR assay with an input of 5 ng cDNA was performed using SsoAdvanced™ Universal SYBR® Green Supermix and SYBR GreenER® qPCR Supermix Universal with and without Eagle’s minimum essential medium (EMEM) supplemented with 10% fetal bovine serum. 10, 20, and 40% EMEM supplemented with 10% fetal bovine serum was spiked in the cDNA to act as an inhibitor. SsoAdvanced Universal SYBR® Green Supermix showed less Cq delay. Cq, quantification cycle.



GC-Rich and AT-Rich qPCR

Excellent qPCR Efficiency and Sensitivity Even with Difficult Target Sequences

qPCR reactions were performed with GC- and AT-rich assays using SsoAdvanced™ Universal SYBR® Green Supermix. Great linearity and efficiency was achieved in both assays, indicating exceptional performance even with challenging assays.

61% GC-rich TIP assay with an input of 50 ng–5 pg gDNA produced standard curve R2 = 0.997, efficiency = 102%, and slope = –3.27



71% AT-rich ADAR sequence with an input of 10 ng–10 pg cDNA produced standard curve R2 = 0.999, efficiency = 92%, and slope = –3.52.



PrimePCR Assay Compatibility

Validated to Generate Best-Quality Data with a PrimePCR Assay

A PrimePCR assay targeting β2 microglobulin with an input of 100 ng–1 pg gDNA was performed using SsoAdvanced™ Universal SYBR® Green Supermix. Results demonstrated outstanding reproducibility and sensitivity over six orders of magnitude. RFU, relative fluorescence units.

Maximum PCR Sensitivity and Specificity

A PrimePCR™ Assay targeting β2 microglobulin, using 250, 125, 62.5, 31.25, 15.625, or 7.8125 cells per 20 µl reaction was performed using SsoAdvanced™ Universal SYBR® Green Supermix. Results show a linear response to varying target concentrations, indicating reliable and effective discrimination of twofold dilutions.



A melt curve analysis of the above reactions shows uniform curves with single peaks, confirming the absence of nonspecific PCR products. RFU, relative fluorescence units.
