PCR inhibitor tolerance testing with SsoAdvanced™ Universal Inhibitor-Tolerant SYBR® Green Supermix against Quanta's PerfeCTa SYBR® Green FastMix and Clontech's Terra qPCR Direct SYBR® Premix. Each qPCR reaction was performed using a challenging long (226 bp) human genomic DNA assay targeting GAPDH and 5 ng of human gDNA. The values represent the maximum percentage and concentration for which a ≤1 Cq delay was noted. Any delay >1 Cq was deemed unacceptable for inhibitor tolerance. The values shaded in green represent the highest levels obtained. The values shaded in orange represent reactions whose melt profiles showed more than one peak. Areas shaded in dark gray were not tested.

Crude lysate tolerance testing with SsoAdvanced™ Universal Inhibitor-Tolerant SYBR® Green Supermix and several competitive SYBR® Green master mixes. Each qPCR reaction was performed using sample-specific qPCR assays. Commercially available sample-specific lysis kits were used to obtain the crude lysates. The values represent the maximum percentage of crude lysate for which a <1 Cq delay was noted. Any delay <1 Cq was deemed unacceptable for inhibitor tolerance. The values shaded in green represent the highest levels obtained. The values shaded in orange represent reactions whose melt profiles showed more than one peak. Comparison of SsoAdvanced™ Universal Inhibitor-Tolerant SYBR® Green Supermix and competitor kit for allele-specific qPCR. Genotyping using an allele-specific SNP (NQ01*2; 43.8% GC, 144 bases) in crude cell lysates from HeLa (wild-type), HCT15 (heterozygote), and HTB26 (mutant) cells.Top 3, Bio-Rad supermix displays high precision, earlier Cq values, and correct genotypes; Bottom 3, competitor mix shows delayed Cq values, low precision, and amplification of the wild-type target in the mutant sample, indicating poor specificity. SsoAdvanced Universal Inhibitor-Tolerant SYBR® Green Supermix is a high-performance real-time PCR supermix based on Bio-Rad's patented* Sso7d fusion protein technology. This supermix is specifically formulated for use with difficult target sequences in a wide range of challenging sample types including crude lysates. The dsDNA-binding protein, Sso7d, stabilizes the polymerase-template complex, providing superior PCR inhibitor tolerance, increased processivity and specificity, and greater speed without affecting PCR sensitivity, efficiency, or reproducibility.
