Effect of Flow Rate

Effect of flow rate on separation and peak symmetry when using Macro-Prep High S media. A 5 ml sample (1.4 mg/ml) of myoglobin (peak 1), ribonuclease A (peak 2), and cytochrome c (peak 3) was run on 1 x 13 cm (8.7 ml) column at each of the indicated flow rates. The buffer was 20 mM potassium phosphate, pH 8.0 with a gradient of 0–1 M KCI over 75 ml.

Macro-Prep High S strong cation exchange support contains sulfonate functional groups and is ideal for purifying basic and neutral proteins and peptides. It is an excellent choice for rapid purification that meets the demands of analytical, semipreparative, and process-scale applications.

This methacrylate copolymer bead provides high-resolution separations at very high flow rates. Changes in pH or ionic strength of the buffer do not cause shrinking or swelling of the support. The superior mechanical and chemical stability of Macro-Prep High S support make it a preferred choice over other sulfonate cation exchangers on the market. Macro-Prep High S can be sanitized with chaotropic agents and can be autoclaved. It provides consistent and reproducible results.
