Quantitation and Precision

iQ™ SYBR® Green Supermix generates precise, quantitative results.

A 5-fold dilution series (50 ng–80 pg) of human genomic DNA was amplified using the supermix and primers and a probe specific to the IL-1β gene. Triplicate reactions at each concentration were amplified along with no-template controls on the iCycler iQ® Real-Time System. r = 0.999, slope = –3.380, efficiency = 97.6%, RFU, relative fluorescence units.



Broad Dynamic Range

iQ™ SYBR® Green Supermix and the iScript™ cDNA synthesis kit show high specificity over a broad dynamic range of cDNA.

Serial dilutions (1 µg–1 pg) of HeLa total RNA were reverse transcribed, and the resulting cDNA was amplified using primers specific to the β-actin gene. Triplicate reactions at each concentration were amplified along with no-template controls on the iCycler iQ® Real-Time System. The consistent spacing of the curves reflects accurate reverse transcription and amplification. r = 0.997, slope = -3.615, efficiency = 89.1%, RFU, relative fluorescence units.



Specificity With Plasmid DNA

iQ™ SYBR® Green Supermix shows high specificity with plasmid templates.

Tenfold serial dilutions (108 to 102 copies) of plasmid DNA were amplified using primers specific to the β-actin gene. Triplicate reactions at each concentration were amplified along with no-template controls on the iCycler iQ® Real-Time System. r = 1.000, slope = -3.492, efficiency = 93.4%, RFU, relative fluorescence units.

SsoAdvanced Universal Supermixes

iQ™ SYBR® Green Supermix is formulated to achieve optimal results for a wide array of real-time PCR applications with the convenience of SYBR® Green I detection chemistry. This preblended 2x solution contains an optimized concentration of SYBR® Green I dye for sensitive and accurate detection, hot-start iTaq™ DNA Polymerase, a proprietary buffer and stabilizers specifically optimized for qPCR using SYBR® Green I, and qPCR-qualified dNTPs. This supermix also contains fluorescein, which is used as a passive reference dye for well-factor collection on certain real-time PCR detection systems. Use of iQ™ SYBR® Green Supermix minimizes preparation time, produces reliable results for real-time PCR assays, and provides precise linear detection over six orders of magnitude.
