Overview

Data Using the Gene Pulser MXcell™ System

Optimization of electroporation conditions. Human primary fibroblast cells denoted as FS (foreskin) were used to compare a square waveform with varying voltages (top) to an exponential waveform with varying capacitance values (bottom) in a 96-well plate. Although the exponential waveform resulted in good data overall, the square waveform resulted in the highest transfection efficiency and cell viability when compared side by side on the same plate. Data Using the Gene Pulser Xcell™ System

Gene Pulser® electroporation buffer produces optimal results in various electroporation systems. Gene Pulser Xcell™, Gene Pulser II, and a competitor's electroporation instrument were used to demonstrate the Gene Pulser electroporation buffer's flexibility. CHO cells were transfected with a scrambled siRNA (green) or an siRNA targeting luciferase (blue). Similar transfection efficiencies were observed across platforms for siRNA delivery.

Gene Pulser electroporation buffer outperforms buffers traditionally used for electroporation when using either square or exponential waveforms. Transfection conditions were optimized in different cell types. For COS-7 cells (top), a square-wave protocol was optimal, whereas for K562 suspension cells (bottom) an exponential-decay protocol was optimal. Under these optimized conditions, Gene Pulser electroporation buffer yielded significantly higher transfection efficiencies than phosphate buffered saline (PBS) and Opti-MEM medium (Invitrogen Corporation), both of which are commonly used for electroporation. Cell Survival

Cell survival under a range of electroporation conditions. Post-electroporation survival of neutrophils was assayed after 24 hr in culture by counting the number of intact cells that excluded trypan blue, and dividing by the number of neutrophils initially transfected. Cell survival was substantially reduced at 300 V and 1,000 µF.

Gene Pulser electroporation buffer is formulated to improve electroporation by minimizing cell mortality while ensuring highly efficient delivery of nucleic acids. This universal buffer is an excellent tool for electroporating mammalian cells, including primary cells and difficult-to-transfect cells.
