Question:

Can I use one channel at a time for ligand immobilization and reserve the blank channels for a future experiment? Moreover, can I reuse a channel after a ligand has been immobilized to the surface of the sensor chip?


Answer:

Yes, it is possible to immobilize ligands in individual channels with any of the amine coupling, histidine-tag binding, and lipid assembly capturing sensor chips. It is also possible to reuse a channel after a ligand has been immobilized to the surface of the sensor chip.

Individual channels on an NLC chip (for biotinylated targets) should not be reused due to the chemistry involved in the stabilization of the NeutrAvidin surface. Each channel should be used only once.
