Question:

When I am using the EpiQ™ Chromatin Analyis Kit, I find that the genomic DNA concentration in the nuclease-treated samples is higher than the untreated samples. What might cause this?


Answer:

It is common for the nuclease-treated samples to have a higher genomic DNA (gDNA) concentration compared to the untreated samples. Smaller DNA fragments elute from the purification column more efficiently than do larger fragments of DNA. Fragments from nuclease-treated samples will be much smaller than fragments from untreated samples and will therefore elute more efficiently and give a higher concentration. This difference in elution efficiency does not affect the outcome of the assay.
