Can I quantitate the proteins on my 2-D gels with SYPRO Ruby protein gel stain?

You can get the relative quantitation of proteins on your 2-D gels, but you cannot get the absolute amount of protein in a specific spot unless you use as a standard the same protein as in the spot.

SYPRO Ruby Protein Gel Stain (1703126, 1703138 and 1703138) has the same staining characteristic of all protein stains; it stains proteins differentially. It binds to some proteins in higher concentration than to other proteins, and exactly how much it binds to a particular protein will vary from protein to protein. The amount that binds must be determined for each protein, and a standard curve for quantitation of a protein is valid only if the same protein is used for the standard as is quantitated in the unknown.

SYPRO Ruby Protein Gel Stain is exceptional in the linearity of fluorescence emitted with increasing amounts of a protein over 3 orders of magnitude. See also Bulletin 2533.
