Question:

All we have is a UV lightbox for visualizing ethidium bromide-stained gels. Is this adequate for crosslinking nucleic acids to membranes after transfer?


Answer:

5 minutes of exposure to a short-wave (254-312 nm) UV light source such as a handheld UV lamp or a UV transilluminator is the next best thing to using a UV irradiation chamber to deliver 200-500 mJ/cm2. While a time course was not run, several wavelengths were tested and showed essentially equivalent signal quality.
