Question:

Can I use DTT and BME with the DC ™ Protein Assay?


Answer:

The standard protocol for the DC Protein Assay cannot tolerate BME and can tolerate only 1 mM DTT (1610611). However, Bulletin 1909 contains a protocol that allows up to 50 mM BME and 5 mM DTT with the DC Protein Assay.

One remedy is to precipitate the proteins with 10% or 12% TCA, resuspend them in buffer without BME or DTT to remove reducing agents, and then continue with the DC protein assay protocol (add Reagent A').
