Question:

I am trying to cast IEF gels at a pH range of 8 or above. The gel is not polymerizing very well, and it is not sticking to the gel support film. What is wrong?


Answer:

Basic ampholytes (pH >8) may interfere with the adhesion of gels to the gel support film. Increasing the ammonium persulfate concentration in the final gel to 0.7 mg/ml should alleviate the problem. Leave the TEMED (1610800) concentration and the flavin concentration the same.

Bio-Rad offers a technical bulletin on acrylamide polymerization, Bulletin 1156, which has useful information on the correct procedures for good, reproducible polymerization of IEF gels.
