Why does Bio-Rad suggest the use of TAE buffer in the DCode™ Cell?


Bio-Rad suggests use of TBE buffer in SSCP gels, which run cold, and TAE buffer in DDGE, TTGE, and CDGE, which run at a higher temperature. TBE has a lower conductivity, and therefore a lower current and potentially a lower temperature than TAE.

Each procedure is validated with the appropriate buffer, as recommended in the manual. Please see also Bulletin 2408, Temporal Temperature Gradient Electrophoresis: A Powerful Technique to Screen Mutations. This bulletin shows data obtained using TAE in the TTGE application, and contains a section on buffer concentration which may be of interest.
