A wide selection of ion exchange media is available in various particle sizes, ionic forms, and purity ranges to offer a variety of purification solutions for process applications.
Binding of polyclonal human IgG by Nuvia S media. Comparison of Nuvia S and other commercially available CEX media. Column size, 0.7 x 5.5 cm. The sample was loaded onto the column in 40 mM sodium acetate, pH 5.0 and 30 mM sodium chloride, washed, and eluted with 40 mM sodium acetate, pH 5.0 and 1 M sodium chloride. DBC, Dynamic Binding Capacity. BT, Breakthrough Ultra-high binding capacities provide users with the maximum value through reduced media and buffer consumption, when used in both capture and polish mode.

High-throughput experimentation (HTE) was used in this case study in concert with design of experiments (DOE) methodology to study the mass recovery of a basic therapeutic protein (pI 9.2–9.5) using a hydrophobic cation exchange mixed-mode resin.
* 10% breakthrough capacity determined with 4.5 mg/ml human IgG (UNOsphere S) and 2.0 mg/ml BSA (UNOsphere Q) in a 1.1 x 20 cm column. ** UNOsphere packed into a 20 cm bed height and run at 1,200 cm/hr generates less than 2 bar backpressure.
Achieve High Productivity Using UNOsphere S Cation Exchange Support
Be Productive In the bioprocess industry, the isolation of biomolecules from crude feedstock is one of the most demanding chromatographic steps in the downstream process. Biopharmaceutical manufacturers are under increasing economic pressure to reduce drug production costs. These factors require the media used in the capture step to have very high binding capacities at fast linear velocities while maintaining low column backpressure. UNOsphere is a patented* new-generation polymeric support, based on a single-step polymerization process that delivers high productivity in the capture step.

Purification of a Monoclonal Antibody Using a Combination of UNOsphere Rapid S and UNOsphere Q Media Ion Exchange Chromatography After Protein A Capture
Introduction Antibody-based drugs represent a growing segment of the pharmaceutical industry and one of the most promising classes of therapeutic drugs. The demand for protein-based drugs is expected to grow steadily for the foreseeable future. However, production of monoclonal antibody (mAb)–based drugs remains very costly, and solutions to lower production costs are needed. Downstream purification steps are becoming a target for cost reduction. The use of ion exchange chromatography to directly capture MAbs from cell culture streams or as a polishing step after an affinity separation may be a way to reduce costs.
Achieve High Productivity Using Macro-Prep High S Cation Exchange Support
Summary Ion exchange chromatography is one of the most widely used techniques for protein purification. Macro-Prep High S support is a strong cation exchanger containing sulfonate functional groups and is ideal for purifying basic and neutral proteins and peptides. Macro-Prep High S support is an excellent choice for rapid purification. This methacrylate copolymer bead provides high-resolution separations at very high flow rates. Changes in pH or ionic strength of the buffer do not cause shrinking or swelling of the support. The superior mechanical and chemical stability of Macro-Prep High S support make it a preferred choice over other sulfonate cation exchangers on the market.
