HeLa cells were transfected with GFP and sorted by forward and side scatter, singlets, and GFP expression. A–B, presort analysis of GFP-positive cells; C–D, postsort population was sorted to >98% purity. Postsort analysis revealed that a pure GFP-positive population was successfully sorted.

SPHERO rainbow calibration particles (8 peaks) were excited by both 488 nm and 561 nm lasers. Emission was measured by all four detectors. Between peaks, all channels displayed a coefficient of variation (CV) <2.5 units. The FL3 channel displays the seven lowest peaks. The eighth peak is offscale due to the brightness of the signal intensity.

Carryover between samples is minimized using the “lock and load” backflushing system. FITC-labeled beads were run on the instrument (A). Subsequently, PE-labeled beads were run (B), followed by a repeated cycle of FITC-labeled beads (C) and PE-labeled beads (D). Each time, carryover of fluorescently labeled beads was analyzed and showed minimal carryover between samples.

Optimal Cell Sorting Performance
and Purity

Obtaining optimal sort purity and maximizing recovery requires accurate drop delay determination, or measurement of the distance between the laser point of analysis and the last connected drop.

Historically, the drop delay process is labor intensive and requires bead counting on a microscope or image processing with a camera.

ProDrop™ technology automates drop delay determination and accurately calculates the drop delay value, resulting in less opportunity for error and leading to better recovery. The exact drop delay value is calculated by measuring the number of ProLine™ calibration beads in the waste stream during the QC process.

Easy access to the S3 cell sorter

The S3 cell sorter is an automated benchtop cell sorter featuring either one or two lasers and up to four fluorescence detectors plus forward- and side-scatter detectors. The S3 cell sorter uses state-of-the-art ProDrop™ technology for automated drop delay calculation and droplet break-off monitoring, thereby simplifying one of the most complex and error-prone aspects of cell sorting.

This affordable cell sorter provides exceptional sort purity without compromising performance and sensitivity. The system is ideal for sorting cells expressing fluorescent proteins such as eGFP, DsRed, mCherry, or for cells labeled with fluorescent markers such as labeled antibodies.

For added biosafety during the cell sorting process, the optional S3 biosafety system Class I (145-1078) is available. The biosafety system is an affordable alternative to the large and expensive traditional biosafety hoods offered for cell sorters.
