Exclusive Sso7d Fusion Polymerase Technology to Empower Your PCR

Sso7d, a 7 kD protein obtained from Sulfolobus solfataricus, binds to dsDNA without any sequence specificity.

Sso7d is covalently linked to a DNA polymerase and is able to stabilize the polymerase-template complex without compromising the structural integrity, thermal stability, or catalytic activity of the enzyme.

The unique structure of this Sso7d fusion polymerase provides several significant advantages over ordinary DNA polymerase:

Obtaining enough starting material to complete multiple gene expression experiments can be challenging when working with precious and limited samples. Optimal Preamplification of Low cDNA Input

SsoAdvanced™ PreAmp Supermix provides optimal preamplification of low cDNA input.

NANOG (A, a high expressor), PAX6 (B, a medium expressor), and NEUROG2 (C, a low expressor) show consistent expression levels regardless of the cDNA input (10 ng to 100 pg). cDNA was prepared from a panel of 22 neuronal stem cells treated with retinoic acid for up to 7 days. 1 µg cDNA was used for the no preamplification control.

SsoAdvanced™ PreAmp Supermix is a 2x concentrated, ready-to-use reaction master mix optimized for unbiased, target-specific preamplification of cDNA and genomic DNA. This PreAmp Supermix preamplifies DNA templates using up to 100 PrimePCR™ PreAmp SYBR® Green or probe-based qPCR assays, custom-designed SYBR® Green and probe-based assays (5’ nuclease), and TaqMan Assays.

Preamplification of Targets without Bias Is Key

To obtain valid real-time PCR and end-point PCR data from limited samples, preamplification is often required. Some methods have been shown to introduce bias across the transcriptome/genome when comparing preamplified and nonpreamplified samples. The SsoAdvanced PreAmp Supermix is the first supermix that truly provides unbiased preamplification reactions regardless of the targets present in the reaction master mix, enabling more data to be obtained from a limited source.

Setting up a preamplification takes only a few minutes. Simply combine the pooled assays, SsoAdvanced PreAmp Supermix, and template in a microcentrifuge tube, and start the PCR reaction.
