Hard-Shell® PCR Plate Compatibility Chart

Hard-Shell PCR plates represent an innovative concept in PCR plate design. The skirt and deck are molded from a rigid thermostable polymer. This hard shell prevents the distortion and shrinkage that may occur when regular single-component polypropylene PCR plates are exposed to the high temperatures of thermal cycling or heat sealing. Bio-Rad offers a variety of Hard-Shell 96-well and 384-well PCR plates.

Two-Component Design for Superior Uniformity and Precise Handling

Hard-Shell PCR plates represent an innovative concept in PCR plate design. The skirt and deck are molded from a rigid thermostable polymer. This hard shell prevents the distortion and shrinkage that may occur when regular single-component polypropylene PCR plates are exposed to the high temperatures of thermal cycling or heat sealing. In a separate step, the thin-wall wells are molded of virgin polypropylene selected for low DNA binding. The footprint and well spacing of Hard-Shell skirted PCR plates meet ANSI/SBS microplate standards, and raised rims around each well allow tight sealing with a wide variety of sealing systems, including adhesive-, pressure-, and heat-sealing methods. Hard-Shell PCR plate attributes include:


  • Superior stability and flatness due to a patented* 2-component design, allowing precise positioning for automation
  • Extremely uniform wells that reduce well-to-well variability in optical assays, such as those performed in real-time PCR
  • Thin wall for fast PCR
  • Several plate styles that fit most thermal cyclers, real-time PCR detection systems, DNA sequencers, and plate readers
  • Black lettering for easy well identification
  • White-well option, which allows increased fluorescent signal strength
  • Color-coded skirts with clear wells

Well Color Effect on Signal Strength

White well:

  • Increases fluorescent signal for maximizing detection sensitivity
  • Prevents fluorescent interference from adjacent wells or a contaminated cycler

Clear well:

  • Transparent wells for easy sample handling
  • Low signal variability for consistent results

Black well:

  • Lower signal strength for FP (fluorescence polarization) and other assays where reflected light must be minimized

Optical flat cap strips:

  • Vapor-tight sealing for tube strips or 96-well plates
  • Suitable for PCR volumes >5 µl

Microseal® 'B' adhesive seals, optically clear:

  • Higher optical transmission than optical caps
  • Suitable for PCR volumes >10 µl

Clear Chill-out™ liquid wax:

High-Profile vs. Low-Profile

 

There are two common well heights for PCR tubes and plates:

  1. High-profile (full-height; around 20.7 mm) wells are “standard” and fit in most classical thermal cyclers and real-time PCR detection systems, such as Bio-Rad's iCycler®, iQ™ series real-time PCR detection systems, and Applied Biosystems' regular PCR systems and DNA sequencers.
  2. Low-profile (around 15.5 mm) wells are the newer design that reduces the potential for condensate formation and offers advantages for light capture in fluorescence assays, low-volume reactions, and fast PCR. They have been recommended for newer instruments such as Bio-Rad's C1000 Touch™ and S1000™ thermal cyclers, CFX series real-time PCR detection systems, and Applied Biosystems' fast PCR systems.

Instruments with adjustable lids can usually use both high-profile and low-profile wells. These instruments include Bio-Rad's 1000-series and DNA Engine® series thermal cyclers.

Hard-Shell low-profile 96-well semi-skirted PCR plates are compatible with many cyclers, including Applied Biosystems 7500 fast and ViiA 7 fast instruments and many of Bio-Rad’s thermal cyclers and real-time PCR detection systems. Hard-Shell plates are specifically designed to withstand the stresses of heat sealing, thermal cycling, and robotic handling. The patented* two-component microplate design provides superior stability and flatness, allowing precise positioning for automation. The low-profile wells are optimized for low-volume reactions and fast PCR.
